实验原理
本试剂盒应用双抗体夹心酶标免疫分析法测定标本中hs-CRP水平。用纯化的抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入hs-CRP抗原、生物素化的抗人hs-CRP抗体、HRP标记的亲和素，经过彻底洗涤后用底物TMB显色。TMB在过氧化物酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的hs-CRP呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），计算样品浓度。

Intended use
This immunoassay kit allows for the specific measurement of Human High sensitivity C-reactive protein (hs-CRP) concentrations in cell culture supernates, serum and plasma.

Introduction
C-reactive protein (CRP) is a liver-derived pattern recognition molecule that is increased in inflammatory states. It rapidly increases within hours after tissue injury, and it is suggested that it is part of the innate immune system and contributes to host defense. Since cardiovascular disease is at least in part an inflammatory process, CRP has been investigated in the context of arteriosclerosis and subsequent vascular disorders. Based on multiple epidemiological and intervention studies, minor CRP elevation [high-sensitivity CRP (hsCRP)] has been shown to be associated with future major cardiovascular risk.CRP is known for several decades as a nonspecific inflammation marker. High CRP levels are detected in human blood during bacterial, viral and other infections, as well as in noninfectious diseases such as rheumatic disorders and malignancies. In acute coronary syndromes raised concentrations of CRP may be a response to myocardial necrosis. In the patients with unstable coronary disease the concentration of CRP changes from 4 to 26 mg/l. Elevated level of CRP at the time of hospital admission of the patients with unstable angina is a predictive value for future ischemic events. CRP measurements in the patients with ischemic heart disease provide a novel method for detecting individuals at high risk of plaque rapture.However, only high-sensitivity (hsCRP) or ultra-sensitive tests for CRP are useful for predicting heart attacks.

Test principle
This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for hs-CRP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any hs-CRP present is bound by the immobilized antibody. An enzyme-linked monoclonal antibody specific for hs-CRP is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of hs-CRP bound in the initial step. The color development is stopped and the intensity of the color is measured.
